This medication is a tricyclic antidepressant, prescribed for depression It is also prescribed for migraine and nerve pain It corrects the imbalance of chemical substances acting on the nervous system


Mechanism of Action
  • Amitriptyline is a dibenzocycloheptadiene tricyclic antidepressant
  • It increases synaptic concentration of serotonin and/or norepinephrine in the CNS by blocking the neuronal reuptake of norepinephrine and serotonin

Dosage
Depression- Initial: 50-75 mg/day; up to 150 mg/day if needed. Max: 300 mg/day in severe cases Neuropathic pain- Initial: 10-25 mg/day at night, up to 75 mg/day if needed Migraine prophylaxis- Initial: 10 mg/day at night

Warnings/Precautions
Not intended for treatment of bipolar depression. Avoid abrupt withdrawal Patient at risk of seizures, w/ DM, narrow angle glaucoma, urinary retention, prostatic hyperplasia, chronic constipation, history of CV disease Renal and hepatic impairment

Mechanism of Action

It works by enhancing the action of a signaling chemical called gamma aminobutyric acid (GABA) in the brain. This results in a calming effect on the nerves and helps to reduce nervous tension (anxiety) and muscle spasm


Dosage
The recommended dose is 20 to 100mg

Side Effects
Mouth dryness, blurring, hesitancy and urgency in urination, constipation, fever, heat stroke, fast heart rate and dilatation of pupils Headache, nervousness, drowsiness and weakness Nausea, aggression and mild skin rashes

Warnings/Precautions
Caution should be exercised in patients with history of heart disease, congestive heart failure, hernia, lung or breathing problems [COPD], nerve and muscle problems, prostate problems, high blood pressure, thyroid, kidney, heart, or liver disease, any allergy, who are taking other medications, elderly, children, during pregnancy and breast feeding It may cause drowsiness, do not drive a car or operate machinery while taking this medication Avoid alcohol consumption Monitor liver function and complete blood cell counts regularly while taking this medication
