DNA is the hereditary element of humans. It consists of strands made of sugar and phosphate. Scientifically, DNA strands are attached with four types of molecules or bases, which contain the information on DNA. DNA Fragmentation is an anomaly that occurs when alteration to these bases takes place. In other words DNA Fragmentation refers to a physical breakdown of one or both DNA strands.The genetic integrity of the sperm is crucial for successful fertilization and normal embryo development. Sperm DNA fragmentation is a term used to denote when the genetic material within the sperm is abnormal, which in turn may lead to male subfertility and IVF failure. Often, men with problems in sperm motility or abnormal sperm morphology faces sperm DNA fragmentation issue, due to physical collapse of one or both DNA strands in sperm chromosomes.

As part of DNA Fragmentation test the collected semen sample of the Male partner is sent to Sperm DNA fragmentation Centre for testing. The semen sample is tested through technique of Sperm Chromatin (SCD), Tunel Assay, Commet Assay and Sperm Chromatin Structure Assay. Based on these tests diagnostic reports are prepared and shared with the patient.

  1. A couple can conceive if the male has issues with DNA fragmentation. However, if the percentage of sperm carrying high rates of DNA fragmentation is higher than normal (above 30%); the rate of conception is much lower.
  2. Even if pregnancy occurs with fragmented DNA, the embryo/s formed from such DNA faces the issue of prognosis (risk with survival), formation of abnormal mutations during blastocyst stage of the embryo, miscarriage and abnormalities in the child born with potential of future cancer risks.

  5. DNA Fragmentation test is necessary when female partner is facing recurrent miscarriages for a long time.

Sperms with DNA fragmentation has the ability to fertilize although there is a risk with the offspring born or the embryo formed out of it. Some of the viable treatments are as follows.

  1. Use of anti-biotics in the infected area especially pelvic area to help protect from future trouble such as growth of varicocele (abnormalities in the veins within the scrotum).
  2. Changes in life style including a ‘Strict No’ to drugs, alcohol and smoking. Also diet rich food with antioxidants and vitamin C and E are very beneficial.
  3. Use of sperm retrieval methods such as TESA (Testicular Sperm Aspiration) to treat infertility as artificially retrieved sperm has better DNA integration than naturally ones.
